Okay, a couple of straggler comments. You are designing an assay inside of living cells, so you can't assume that the pH in your cell supernatant is going to change in response to the reaction, nor can you assume that a reaction that releases NH4 will necessarily release a gas into the air outside of the cell, much less one that is detectable by scent. And remember that buffers are designed to suppress changes in pH in the solution.
